(アルキル化BTP-Neu5Acの合成)
本実施例では、上記実施例1で合成したBTP3-Neu5Ac前駆体methyl 2-(benzothiazol-2-yl)-4-bromophenyl-(5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero-D-galacto-2-nonulopyranosid)onateを使用してアルキル化BTP-Neu5Acの合成を行った。詳細は、以下の通りである。なお、アルキル化BTP-Neu5Acのナトリウム塩を最終産物とした。

(アルキル化BTP-Neu5Acの合成方法)
○pC8-ane-BTP-Neu5Acの合成
methyl 2-(benzothiazol-2-yl)-4-bromophenyl-(5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero-D-galacto-2-nonulopyranosid)onate(160 mg, 200 μmol), 1-octyne (59 μl, 400 μmol), CuI (7.6 mg, 40 μmol), PdCl
2(Ph
3P)
2(28 mg, 40 μmol), Et
3N (0.2 mL), CH
3CN (3 mL) の混合物をアルゴン雰囲気下、80℃で15時間撹拌した。放冷後、反応液を濃縮し、シリカゲルカラムクロマトグラフィー(シリカゲル: 100 mL、溶媒AcOEt)で精製し、褐色泡状のpC8-yne-BTP-Neu5Ac前駆体 (104 mg, 収率64%) を得た。
得られたpC8-yne-BTP-Neu5Ac前駆体 (81 mg, 100 μmol), 10% Pd/C (10 mg) をMeOH (2 mL) に縣濁し、水素雰囲気下で17時間撹拌した。Pd/Cを濾別して濃縮してpC8-ane-BTP-Neu5Ac前駆体 (87 mg) 得た。得られた前駆体 (60 mg, 73.8 μmol) のMeOH (0.3 mL) 溶液にNaOMe (5 M in MeOH, 30 μL, 150 μmol) を加えて1時間撹拌し、NaOH水溶液 (2.5 M, 295 μL, 738 μmol) を加えてさらに16時間撹拌した。酢酸 (51 μL) を加えて中和後、HPLC(Triart C18, 10 mM NH
3/H
2O/CH
3CN) で精製し、NaOH水溶液で中和して凍結乾燥によりpC8-ane-BTP-Neu5Ac (34.7 mg, 収率71%) を無色不定形物として得た。pC8-ane-BTP-Neu5Ac以外のアルキル化BTP-Neu5Acも同様な方法で合成した。
