2の合成
(国際特許出願第2004/014415号に記載されており、その全体が本明細書において参照により援用される)。以下に記載の方法において、全ての等価物は樹脂−バッチスケールに関連する。

表題のペプチドを50リットルの濾過反応器(Buchi社、スイス フラヴィル)内で合成した。

合成は1.04モルスケール(1.4kgの樹脂を投入)で実施した。

樹脂からのペプチドキメラの切断及び側鎖保護基の除去は、TFA:TIPS:DCM(15:5:80、12体積、34.3L)を用いてワンポットで2時間行った。断続的な窒素ガスの注入を切断反応混合物に用いた(30分ごとに1−2分の持続時間)。樹脂から切断混合物(所望のペプチドを含有する)を濾過した後、「使用済み」の樹脂をTFA/DCM(15:85)混合物(樹脂重量に対して1.3体積、3回)で洗浄した。ペプチドに富んだ濾液を合わせ、元の濾液重量の10.4%(6.2Kg)まで蒸発させた。粗ペプチドを撹拌MtBE(蒸発後の残留物重量に対して6.5体積、40L)に添加することにより濃縮溶液から沈殿させ、濾過してMtBE(蒸発後の残留物重量に対して1体積、6.2L、1回)で洗浄した。その後の環化のために、半乾燥ペプチド沈殿物の溶解を、樹脂重量に対して38体積(45L)のACN(30%v/v)を含む0.1%v/vのTFA/水を用いて実施した。
